Background:

  • Tamm-Horsfall protein (THP) is a glycoprotein produced in the kidney, but its function remains unclear.
  • Previous studies have not fully elucidated the interactions of THP with other biological molecules.

Purpose of the Study:

  • To investigate the interaction between sheep THP (sTHP) and sheep IgG.
  • To determine if THP has a role in immunomodulation.

Main Methods:

  • Sheep THP and IgG were purified.
  • Enzyme-linked immunosorbent assay (ELISA) was used to assess binding affinity between sTHP and sheep IgG.
  • Human THP (hTHP) and human IgG interaction was also examined.

  • A high molecular weight contaminant in sTHP preparations was identified as sheep IgG.
  • ELISA revealed high and low affinity binding sites between sTHP and sheep IgG (Kd 10⁻⁸–10⁻⁹ M and 10⁻⁶–10⁻⁷ M, respectively).
  • Similar high affinity interactions were observed between hTHP and human IgG, and soluble sTHP inhibited IgG binding.
  • Conclusions:

    • Tamm-Horsfall protein physically interacts with immunoglobulin G in both sheep and humans.
    • These findings suggest a potential immunoregulatory function for THP that warrants further research.
