    Summary

    Intravenous nicotine significantly increases plasma arginine vasopressin (AVP) levels and causes behavioral changes in conscious monkeys. Antihistamine pretreatment reduced these nicotine-induced effects.

    Background:

    • Arginine vasopressin (AVP) plays a crucial role in regulating physiological functions.
    • Nicotine is a widely consumed psychoactive substance with known physiological effects.

    Purpose of the Study:

    • To investigate the impact of intravenous nicotine administration on plasma arginine vasopressin (AVP) levels.
    • To assess nicotine's effects on plasma osmolality and observable behaviors in conscious monkeys.

    Main Methods:

    • Adult female rhesus monkeys (Macaca mulatta) received intravenous infusions.
    • Control experiments involved dextrose and water infusion.
    • Nicotine infusion (100 mug/kg/min) was administered, with some subjects pretreated with antihistamines (promethazine, diphenhydramine).

    Main Results:

    • Nicotine infusion significantly elevated plasma AVP levels from 0.6 +/- 0.5 muU/ml to 35 +/- 17 muU/ml.
    • Behavioral changes including restlessness, yawning, retching, salivation, and chewing were observed during nicotine infusion.
    • Plasma osmolality remained unchanged, and antihistamine pretreatment attenuated both AVP increase and behavioral effects.

    Conclusions:

    • Nicotine administration triggers a substantial release of arginine vasopressin in conscious monkeys.
    • Nicotine-induced AVP release and associated behaviors are, in part, mediated by histamine pathways.
