Summary

Lesioning the Nucleus of the Optic Tract (NOT) and Dorsal Terminal Nucleus (DTN) in monkeys impaired optokinetic nystagmus (OKN) and smooth pursuit eye movements, revealing a link between these systems at the pretectal level.

Area of Science:

  • Neuroscience
  • Ophthalmology
  • Vision Science

Background:

  • The Nucleus of the Optic Tract (NOT) and Dorsal Terminal Nucleus (DTN) are crucial for horizontal optokinetic nystagmus (OKN).
  • Previous research on NOT/DTN cells in rhesus monkeys provided single-unit data, but the link between pursuit and optokinetic systems at this level remained unclear.

Purpose of the Study:

  • To investigate the role of the NOT/DTN in both optokinetic and pursuit eye movements.
  • To determine if the NOT/DTN is essential for integrating visual motion information for different oculomotor behaviors.

Main Methods:

  • An electrolytic lesion was strategically placed in the NOT/DTN region of rhesus monkeys.
  • Monkeys were subsequently tested using both optokinetic and smooth pursuit eye movement paradigms before and after the lesion.

Related Experiment Videos

Main Results:

  • Immediately post-lesion, a spontaneous nystagmus with slow phases directed away from the lesioned side was observed, persisting even during contra-lesional optokinetic stimulation.
  • While OKN recovery occurred within a week, its gain was significantly reduced for ipsi-lesional stimuli.
  • Smooth pursuit showed a directional deficit, with greater eye velocity reduction when pursuing towards the lesioned side, compensated by increased catch-up saccades.

Conclusions:

  • Lesions of the pretectum, specifically the NOT/DTN, confirm known directional deficits in the optokinetic system.
  • This study demonstrates a novel directional deficit in the smooth pursuit system following NOT/DTN lesions, indicating its involvement in both visual tracking systems.
