Purpose of the Study:

  • To analyze the complement activity of normal human corneal tissue.

  • Human corneal extract (HCE) was prepared and its complement (C) activity assessed via hemolytic assay.
  • Anion-exchange chromatography and gel filtration were used for biochemical analysis.
  • Monoclonal anti-human C3 antibodies were employed for C3 detection and removal.

Main Results:

  • HCE exhibited low intrinsic C-activity (<200 U/g tissue).
  • Unexpectedly, HCE significantly increased serum C-activity in a dose-dependent manner.
  • This enhancing effect was abolished by heat treatment (56°C) and linked to two protein peaks, one identified as C3.

Conclusions:

  • The human cornea possesses an unidentified, heat-sensitive factor that enhances serum complement activity.
  • This factor may play a crucial role in both normal corneal function and disease states.
