    Background:

    • The Golgi-Cox method is a traditional technique for visualizing neural structures.
    • Studying superficial neural tissues requires precise and detailed impregnation methods.

    Purpose of the Study:

    • To modify the Golgi-Cox method for enhanced impregnation of goldfish optic tecta.
    • To evaluate the effectiveness of diluted impregnation solutions for neural tissue morphology.

    Main Methods:

    • Goldfish optic tecta were treated with egg yolk and immersed in modified Golgi-Cox impregnation fluids (solutions A+B diluted).
    • Tecta underwent further processing including immersion in LiOH/KNO3, acetic acid wash, dehydration, and embedding in nitrocellulose.
    • Sections were cut at 100 microns and cleared with cedarwood oil for microscopic examination.

    Main Results:

    • Diluted impregnation solutions (methods b and c) resulted in superior cell impregnation compared to the original method (a).
    • Enhanced impregnation was observed in layers 200-400 microns below the surface, with well-preserved dendrites and unmyelinated axons.
    • Myelin sheaths were successfully impregnated and identifiable by their characteristic chain-like appearance.

    Conclusions:

    • The modified Golgi-Cox method provides superior impregnation of neural cells and processes in goldfish optic tecta.
    • This technique is effective for studying the morphology of superficially located nervous tissue, including myelin sheaths.
