Background:

  • Osteocalcin (OC) is a vitamin K-dependent protein specific to bone, but its precise physiological role is not fully understood.
  • Previous studies in rats showed that vitamin K deficiency or warfarin treatment reduces bone OC levels, as noncarboxylated OC does not bind to bone hydroxyapatite.

Purpose of the Study:

  • To investigate the role of osteocalcin in bone remodeling using a warfarin-induced depletion model in growing lambs.
  • To analyze the effects of chronic warfarin and vitamin K1 treatment on bone histomorphometry and osteocalcin levels.

Main Methods:

  • Growing lambs were divided into four groups: control, vitamin K1, vitamin K1 + warfarin (75 mg/kg/day), and vitamin K1 + warfarin (150 mg/kg/day).
  • Treatments were administered daily for 3 months.

  • Bone osteocalcin levels, serum noncarboxylated osteocalcin, and histomorphometric parameters (cancellous bone area, eroded perimeter, bone formation rate) were analyzed.
  • Main Results:

    • Warfarin treatment significantly decreased bone osteocalcin levels in both the metaphysis and diaphysis of metacarpals.
    • Serum noncarboxylated osteocalcin levels were significantly higher in warfarin-treated lambs compared to controls.
    • Histomorphometric analysis revealed significantly reduced cancellous bone area, eroded perimeter, and bone formation rate in warfarin-treated lambs.

    Conclusions:

    • Depletion of osteocalcin in lamb bone over 3 months induced osteopenia characterized by decreased bone resorption and a more pronounced decrease in bone formation.
    • These findings suggest that osteocalcin plays a vital role in maintaining normal bone mass and facilitating trabecular bone remodeling.
