Glomerular protein synthesis in proteinuric rats

    Summary

    Proteinuria in rats, induced by bovine albumin, does not increase glomerular basement membrane (GBM) synthesis. Studies show increased synthesis of various glomerular proteins, not specifically GBM, during induced proteinuria.

    Background:

    • Proteinuria, a condition characterized by abnormal protein levels in urine, can be induced in rats via bovine albumin injection.
    • Previous research indicates that induced proteinuria is associated with glomerular ultrastructural and enzyme alterations.

    Purpose of the Study:

    • To investigate if proteinuria and associated glomerular changes involve increased glomerular protein synthesis.
    • Specifically, to determine if there is an elevated synthesis of the glomerular basement membrane (GBM).

    Main Methods:

    • Young rats were induced into a proteinuric state using bovine albumin injections.
    • Glomeruli were isolated using a sieve technique and incubated with radiolabeled amino acids ([U-14C]proline, [U-14C]leucine, [U-14C]tyrosine).
    • Control incubations were performed with glomeruli from normal rats; subsequent analysis involved protein hydrolysis and amino acid radioactivity measurement.

    Main Results:

    • Proteinuric glomeruli exhibited higher specific activities for all three tested amino acids compared to controls.
    • However, the ratio of 14C-hydroxyproline to 14C-proline remained unchanged between proteinuric and normal glomeruli.
    • The uniform increase in amino acid incorporation suggests a broader increase in cellular protein synthesis.

    Conclusions:

    • The observed increase in amino acid incorporation in proteinuric rat glomeruli reflects a general increase in cellular protein synthesis.
    • This study concludes that induced proteinuria does not lead to a preferential increase in glomerular basement membrane (GBM) synthesis.
