    Human adenovirus type 5 infection causes specific phosphorylation of viral and cellular polypeptides. These include ribosomal, nuclear, and membrane-associated proteins, identified using 32P labeling and SDS-PAGE.

    Background:

    • Human adenovirus type 5 (HAdV5) infection alters cellular processes.
    • Viral infection can induce changes in host cell protein phosphorylation.

    Purpose of the Study:

    • To identify virus-induced phosphorylated polypeptides in HAdV5-infected cells.
    • To characterize the location and timing of these phosphoproteins.

    Main Methods:

    • Cells infected with HAdV5 were labeled with 32P-orthophosphate.
    • Extracts were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and autoradiography.
    • Immunoprecipitation with virus-specific antiserum was performed.

    Main Results:

    • Specific phosphorylation of several polypeptides was detected post-infection.
    • Early infection showed phosphorylation of a 26K ribosomal polypeptide.
    • Later infection revealed 100K (ribosomal) and 39K (nuclear) phosphoproteins.
    • A 72K nuclear phosphoprotein precipitated with antiserum, and an 18K membrane-associated phosphoprotein were identified.
    • The structural viral polypeptide IIIa (66K) was also found to be phosphorylated.

    Conclusions:

    • HAdV5 infection induces specific phosphorylation of multiple viral and cellular proteins.
    • These phosphoproteins are localized in different cellular compartments (nucleus, ribosomes, membranes).
    • Phosphorylation patterns change throughout the infection cycle.
