Summary

Clomiphene citrate significantly increases acrosome reaction in spermatozoa in a dose- and time-dependent manner. This effect impairs sperm motility and viability, and is independent of anti-oestrogenic properties.

Purpose of the Study:

  • To investigate the time- and dose-dependent effects of clomiphene citrate on human spermatozoa.
  • To determine the impact on acrosome reaction, motility, and viability.

Main Methods:

  • Fertile donor spermatozoa were incubated with varying concentrations (1-1000 mumol/l) and durations (1-4 h) of clomiphene citrate.
  • Percentages of acrosome-reacted, motile, and live spermatozoa were assessed.

Main Results:

  • Clomiphene citrate induced a dose- and time-dependent increase in acrosome reaction, reaching up to 100%.
  • Acrosome reaction was associated with decreased sperm motility and viability.
  • Optimal conditions for live, reacted spermatozoa were 100 mumol/l for 1 h (37% +/- 1%).
  • Oestradiol did not affect sperm function or inhibit clomiphene's action.

Conclusions:

  • Clomiphene citrate strongly triggers the acrosome reaction in spermatozoa.

  • Clomiphene citrate impairs sperm motility and viability during the acrosome reaction.
