Summary

Free hemin is toxic to heart cells, causing reduced beating and cell damage. This study shows hemin damages the sarcolemma and cell interior, leading to loss of function in neonatal myocytes.

Background:

Free hemin, an iron-containing porphyrin, is implicated in various pathological conditions.
Understanding hemin's direct effects on cardiac cells is crucial for cardiovascular disease research.

Purpose of the Study:

To investigate the toxic effects of free hemin on neonatal cardiac myocytes.
To elucidate the mechanisms underlying hemin-induced cardiotoxicity.

Main Methods:

Primary cultures of neonatal rat myocytes were exposed to varying concentrations of free hemin (up to 20 microM).
Cellular viability was assessed using the trypan blue exclusion test.
Myocyte beating rate and contractility were monitored over time.
Release of cytosolic enzymes was measured to assess cell damage.
Morphological changes were observed using microscopy.

Main Results:

Hemin exposure led to a time- and concentration-dependent reduction in myocyte beating rate.
Arrhythmic beating, loss of contractility, and significant morphological changes (granulation to cell lysis) were observed.
A biphasic release of cytosolic enzymes indicated sequential damage to the sarcolemma and intracellular components.
Hemin-induced cell death correlated with enzyme release in the initial phase, with greater release in the second phase.

Conclusions:

Free hemin exhibits direct cardiotoxicity in neonatal myocytes.
Hemin-induced damage involves initial sarcolemma disruption followed by prolonged intracellular damage.
These findings highlight hemin's potential role in myocardial injury and dysfunction.
