Purpose of the Study:

  • To investigate in vitro racial differences in lymphocyte beta-receptor sensitivity to beta-blockade.
  • To elucidate potential mechanisms for differing responses to beta-blocker therapy.

  • Isolated lymphocytes from healthy white and black male volunteers.
  • Assessed beta-receptor sensitivity using isoproterenol stimulation with and without propranolol.
  • Measured cyclic AMP (cAMP) levels via radioimmunoassay and analyzed using nonlinear regression.

Conclusions:

  • Racial disparities in beta-2 receptor affinity for propranolol may explain differences in antihypertensive response.
  • Higher propranolol concentrations are needed in black individuals to achieve comparable beta-blockade.
