Perforin-expressing cells during spontaneous abortion

Summary

Granulated metrial gland (GMG) cells, expressing perforin, were similarly distributed near healthy and aborting conceptuses in mice. This suggests GMG cells are not typically involved in spontaneous abortions, though other perforin-positive cells were occasionally observed.

Area of Science:

  • Reproductive immunology
  • Cellular immunology
  • Pregnancy research

Background:

  • The metrial gland in pregnant rodents houses granulated metrial gland (GMG) cells, which are natural killer (NK)-like cells.
  • GMG cells contain perforin, a cytolytic protein, and NK cells have been linked to spontaneous abortions.

Purpose of the Study:

  • To investigate the distribution of perforin-containing cells in the mouse uterus during normal pregnancy and spontaneous abortions.
  • To determine the role of GMG cells in spontaneous abortion models.

Main Methods:

  • Comparative analysis of perforin-positive cell distribution in the uteri of Swiss mice (normal pregnancy) and CBA/J x DBA/2 mice (spontaneous abortion model).
  • Histological examination of uterine tissues near healthy and aborting conceptuses.

Related Experiment Videos

Main Results:

  • The distribution of perforin-positive GMG cells was consistent near both healthy and aborting conceptuses.
  • In approximately 5% of spontaneous abortion cases, small perforin-positive cells were found near the aborting conceptus.

Conclusions:

  • GMG cells do not appear to be the primary cause of spontaneous abortions in this mouse model.
  • The role of the sporadically observed small perforin-positive cells in spontaneous abortions remains undetermined.
