    Background:

    • Nortriptyline is a tricyclic antidepressant.
    • Smoking is known to affect drug metabolism through induction of cytochrome P450 enzymes.
    • The impact of smoking on nortriptyline pharmacokinetics requires further investigation.

    Purpose of the Study:

    • To investigate the effect of cigarette smoking on steady-state plasma concentrations of nortriptyline.
    • To determine if smoking status influences nortriptyline levels in patients.

    Main Methods:

    • A study was conducted with 22 cigarette smokers and 31 nonsmokers.
    • Steady-state plasma levels of nortriptyline were measured.
    • Data analysis considered age, sex, and smoking habits.

    Main Results:

    • No significant difference was observed in mean steady-state nortriptyline concentrations between smokers (191.2 +/- 141.3 ng/ml) and nonsmokers (169.3 +/- 92.4 ng/ml).
    • Factors such as age, sex, and the number of cigarettes smoked did not affect plasma concentrations.
    • Individual variability in nortriptyline levels was noted in both groups.

    Conclusions:

    • Cigarette smoking does not appear to alter steady-state plasma levels of nortriptyline.
    • Clinicians can likely maintain standard nortriptyline dosing regardless of smoking status.
    • Further research may explore other factors influencing nortriptyline pharmacokinetics.
