Background:
- Human interleukin-10 (IL-10) is known to inhibit T-cell proliferation and cytokine production, typically in the presence of monocytes.
- The direct effects of IL-10 on T cells, independent of accessory cells, have not been fully elucidated.
Purpose of the Study:
- To investigate whether human interleukin-10 (IL-10) can directly inhibit T-cell proliferation and cytokine production.
Main Methods:
- Used highly purified peripheral blood T cells (<0.1% CD14+ cells) and a human tetanus toxoid-specific T-cell clone (TM11).
- Stimulated T cells with immobilized OKT3 monoclonal antibody (MoAb) in the absence of monocytes.
- Assessed T-cell proliferation and IL-2 secretion, and the effect of IL-10 neutralization.
