Summary

Thyroid very late antigens (VLA) expression differs between normal and diseased glands. A distinct subset of follicular cells with higher VLA expression is significantly increased in multinodular goiters.

  • Thyroid tissue specimens from normal glands and multinodular goiters were processed into single-cell suspensions.
  • Flow cytometry was employed to analyze cell surface protein expression using antibodies against integrin beta 1 and associated alpha chains (alpha 1, alpha 3, alpha 5, alpha 6).
  • Immunofluorescence was used on tissue sections to localize VLA expression on the cell membrane and observe cellular distribution.
  • Main Results:

    • In normal thyroids, >97% of follicular cells expressed beta 1 integrins, predominantly with alpha 3, and low levels of alpha 1, alpha 5, and alpha 6.
    • A minor (<3%) follicular cell population in normal glands showed 10-fold higher beta 1 intensity, with elevated alpha 1, alpha 5, and alpha 6 expression.
    • This minor subset was significantly expanded in multinodular goiters (10-60% of follicular cells), with VLA-expressing cells forming clusters on the basal membrane.

    Conclusions:

    • A distinct subset of thyroid follicular cells characterized by high expression of specific VLA chains (alpha 1, alpha 5, alpha 6) is identified.
    • The significant increase and clustering of this VLA-expressing cell subset in multinodular goiters suggest a role in goiter pathogenesis.
    • These findings highlight potential VLA-mediated mechanisms in thyroid nodule formation and development.
