Summary

Apomorphine (APO) stimulates neuroendocrine responses like growth hormone (GH) and cortisol (COR) release. Schizophrenia patients showed blunted responses, suggesting dopamine receptor subtypes may be involved in psychosis.

Purpose of the Study:

  • To investigate neuroendocrine responses to apomorphine (APO) in patients with schizophrenia (SCZ), schizoaffective disorder (SAD), and major depressive episode (MDE).
  • To compare these responses with those of healthy controls.

Main Methods:

  • 110 drug-free inpatients (SCZ, SAD, MDE) and 18 controls received subcutaneous apomorphine (0.75 mg) or saline.
  • Growth hormone (GH), adrenocorticotropin (ACTH), cortisol (COR), prolactin (PRL), and thyrotropin (TSH) levels were measured.

  • Apomorphine significantly increased GH, ACTH, and COR, and decreased PRL compared to saline.
  • Schizophrenia and SAD groups exhibited significantly lower ACTH and COR stimulation than controls and MDE patients.

Conclusions:

  • Apomorphine-induced ACTH and cortisol stimulation may involve different pathways than GH stimulation.
  • Blunted neuroendocrine responses in schizophrenia and SAD suggest altered dopamine receptor sensitivity.
  • Findings support the hypothesis that distinct psychosis aspects involve different dopamine receptor subtypes.
