Summary

The paraaortic splanchnopleura (P-Sp) is a newly identified embryonic site that generates multipotent hematopoietic stem cells (HSC). These P-Sp derived HSCs contribute to early blood development before fetal liver colonization.

Area of Science:

  • Developmental biology
  • Hematopoiesis
  • Stem cell research

Context:

  • Vertebrate embryonic development involves sequential hematopoietic tissue activation.
  • Yolk sac (YS) was considered the primary source of hematopoietic stem cells (HSC).
  • Recent findings in birds identified HSC origins in the aortic region.

Purpose:

  • To investigate potential intraembryonic sources of mouse hematopoietic stem cells (HSC).
  • To characterize the hemopoietic potential of the paraaortic splanchnopleura (P-Sp) region.
  • To identify early hemogenic sites in mouse embryos.

Summary:

  • A novel in vitro approach detected multipotent hematopoietic progenitors in the mouse paraaortic splanchnopleura (P-Sp).
  • The P-Sp is the sole intraembryonic site producing progenitors that generate B, T, and myeloid cells.

Related Experiment Videos

  • Hemopoietic precursors emerge in both the yolk sac (YS) and P-Sp around the 10-12 somite stage.
  • Impact:

    • Identifies the P-Sp as a crucial early intraembryonic hemopoietic site in mice.
    • Reveals that P-Sp derived hemogenic sites contribute to hematopoiesis before fetal liver colonization.
    • Advances understanding of the origins and early development of hematopoietic stem cells.
