  • Interleukin-12 (IL-12) is a key cytokine regulating T-helper 1 (Th1) immune responses.
  • Epidermal Langerhans cells (LC) are critical antigen-presenting cells (APCs) initiating cell-mediated immunity.

Purpose of the Study:

  • To investigate the capacity of epidermal Langerhans cells (LC) to produce IL-12.
  • To determine if LC contribute to Th1 immune responses through IL-12 production.

Main Methods:

  • Enrichment and depletion of CD1a+ LC from epidermal cell suspensions.
  • Reverse transcription-PCR (RT-PCR) to detect IL-12 p40 mRNA.
  • Radioimmunoassay (RIA) and flow cytometry to quantify IL-12 p40 protein.
  • Bioassay to assess functional IL-12 (p70) activity.
