Background:

  • Allogeneic bone marrow transplantation (BMT) leverages the graft-versus-leukaemia (GvL) effect mediated by donor lymphocytes.
  • The existence of a similar graft-versus-myeloma (GvM) effect in multiple myeloma (MM) requires further investigation.

Observation:

  • Two patients with relapsed multiple myeloma post-allogeneic BMT received leukocyte infusions from their original marrow donors.
  • These infusions contained a significantly higher number of T cells compared to the initial BMT.

Findings:

  • Both patients achieved complete remission of their multiple myeloma.
  • Complete chimerism was observed, with all peripheral blood cells originating from the donor.
  • Patients experienced graft-versus-host disease, indicating immune activation.
