Summary

Optional sequences in fungal mitochondrial DNA, like intronic reading frames, are mobile and can alter adjacent genes. Their gain and loss drive the evolution of mitochondrial genomes in Podospora fungi.

Area of Science:

  • * Mycology
  • * Molecular Evolution
  • * Genomics

Background:

  • * The mitochondrial genome contains optional sequences, including intronic reading frames.
  • * These sequences are found in filamentous fungi of the genus Podospora.
  • * Their role in mitochondrial genome evolution is not fully understood.

Purpose of the Study:

  • * To investigate the role of optional sequences in the mitochondrial genome of Podospora.
  • * To understand the evolutionary dynamics of intronic reading frames.
  • * To explore their impact on adjacent exon sequences and overall genome evolution.

Main Methods:

  • * Examination of mitochondrial genomes from 23 wild-type Podospora strains.
  • * Analysis of optional sequences, including intronic reading frames (e.g., nad1-i4-orf1, cox1-i7-orf2).

Related Experiment Videos

  • * Construction and analysis of heteroplasmons to study ORF invasiveness.
  • Main Results:

    • * Two intronic reading frames (nad1-i4-orf1, cox1-i7-orf2) were identified.
    • * The presence of these ORFs correlated with nucleotide substitutions in adjacent exons.
    • * These ORFs were found to be highly invasive in heteroplasmic progeny.
    • * A scenario for intronic ORF evolution involving gain and loss was proposed.

    Conclusions:

    • * Group 1 intronic ORFs function as mobile elements.
    • * Transfer and modification of ORFs contribute to modular mitochondrial genome evolution.
    • * Recent ORF gain explains sequence differences within the Podospora genus.
