Summary

Short-term hyperbaric oxygenation (HBO) exposure upregulated lung Na,K-adenosine triphosphate (ATPase) mRNA and activity in rats. This suggests HBO may enhance protective mechanisms against lung injury.

Area of Science:

  • Physiology
  • Cell Biology
  • Biochemistry

Background:

  • Hyperoxia increases lung Na+ transport and Na,K-adenosine triphosphate (ATPase) expression.
  • Hyperbaric oxygenation (HBO) can model hyperoxic cell damage but may also trigger protective responses.
  • The effect of short-term HBO on lung Na,K-ATPase prior to injury is unclear.

Purpose of the Study:

  • To investigate if short-term HBO exposure upregulates lung Na,K-ATPase expression and activity.
  • To determine the temporal effects of HBO on Na,K-ATPase alpha-1 and beta-1 mRNA and activity.

Main Methods:

  • Rats were exposed to intermittent or single 2.5-hour HBO sessions.
  • Na,K-ATPase alpha-1 and beta-1 messenger ribonucleic acid (mRNA) transcript levels were measured.
  • Na,K-ATPase activity was assessed at various time points post-HBO exposure.

Related Experiment Videos

Main Results:

  • HBO exposure increased lung Na,K-ATPase alpha-1 and beta-1 mRNA levels up to fourfold.
  • Na,K-ATPase activity in rat lungs doubled within 2 hours post-HBO and remained elevated for 6 hours.
  • These findings suggest HBO enhances Na,K-ATPase expression and activity.

Conclusions:

  • Short-term HBO exposure upregulates lung Na,K-ATPase.
  • HBO may activate endogenous protective mechanisms in the lungs.
  • Further research is warranted to explore HBO's therapeutic potential in lung injury.
