Background:

  • Insulin's role in metabolic regulation is well-established.
  • Its direct effects on platelet function and cyclic guanosine monophosphate (cGMP) signaling are less understood.
  • Platelets play a critical role in hemostasis and thrombosis.

Purpose of the Study:

  • To investigate the concentration-dependent effects of insulin on platelet aggregation and intraplatelet cGMP levels.
  • To elucidate the underlying mechanisms involving guanylate cyclase and phosphodiesterase activity.

Main Methods:

  • Platelet-rich plasma was incubated with varying concentrations of insulin (0.25-200 nmol/L) for 3 minutes.
  • Platelet aggregation induced by adenosine 5-diphosphate (ADP) was measured.

    • In the presence of phosphodiesterase inhibitors, insulin exhibited anti-aggregating effects even at high concentrations.

    Conclusions:

    • Insulin exerts dual, concentration-dependent effects on platelet function.
    • At physiological levels, insulin promotes anti-aggregation by increasing cGMP.
    • At supraphysiological levels, insulin can promote aggregation by decreasing cGMP, primarily through phosphodiesterase activation.
