    Summary

This study analyzed human beta-globin mRNA sequences. Findings reveal dinucleotide reduplication explains abnormal globin variants Tak and Cranston, which can terminate at an out-of-phase codon.

    Area of Science:

    • Molecular Biology
    • Genetics
    • Biochemistry

    Background:

    • Human beta-globin mRNA (beta-mRNA) is crucial for hemoglobin production.
    • Alterations in beta-globin mRNA can lead to various blood disorders like thalassemia and sickle cell disease.

    Purpose of the Study:

    • To perform sequence analysis on human beta-mRNA from different reticulocyte types.
    • To identify the molecular basis of abnormally long beta-globin variants.

    Main Methods:

    • Preparation of complementary DNA (cDNA) from purified beta-mRNA using RNA-dependent DNA polymerase.
    • Transcription of cDNA into complementary RNA (cRNA) using Escherichia coli RNA polymerase.
    • Detailed sequence analysis of the transcribed cRNA.

    Main Results:

    Related Experiment Videos

    • Over 300 nucleotides of the beta-mRNA coding region were identified.
    • 37 nucleotides were assigned to the 3'-terminal noncoding region.
    • The normal termination codon is UAA, with an out-of-phase UAA triplet 28 nucleotides downstream.
    • Abnormally long beta-globin variants (Tak and Cranston) are attributed to dinucleotide reduplication before the normal termination codon.
    • These variants can utilize the out-of-phase UAA triplet for termination.

    Conclusions:

    • Dinucleotide reduplication is the likely cause of the Tak and Cranston beta-globin variants.
    • An alternative termination codon (out-of-phase UAA) can be used by these abnormal variants.
