Summary

Genetic analysis of interferon-alpha genes reveals lower polymorphism than expected. Most previously identified gene sequences may be rare, nonpolymorphic mutants, potentially from tumor cell lines.

Area of Science:

  • Genetics
  • Immunology
  • Molecular Biology

Background:

  • Genetic polymorphism in interferon type I gene family was previously assumed based on RFLP and sequence data.
  • However, only the IFNA2 locus had been analyzed for gene frequency, revealing only rare alleles.

Purpose of the Study:

  • To investigate the genetic variation of expressed interferon-alpha genes (IFNA1, IFNA2, IFNA10, IFNA13, IFNA14, and IFNA17).
  • To determine the extent of genetic polymorphism in these interferon-alpha genes within a population.

Main Methods:

  • Screening of genomic DNA from blood donors and family material using single-nucleotide primer extension (allele-specific primer extension) of PCR fragments.
  • Utilized nested PCR, restriction analysis, and control sequencing to address sequence similarities.

Related Experiment Videos

Main Results:

  • Interferon-alpha 1 and interferon-alpha 13 variants were common; the LeIF D variant was absent.
  • IFNA2 and IFNA14 loci showed only one sequence variant.
  • IFNA10 and IFNA17 groups each had two alleles, which segregated in families and fit Hardy-Weinberg equilibrium.
  • Significant linkage disequilibrium was observed between IFNA10 and IFNA17 alleles.

Conclusions:

  • The genetic polymorphism of the studied interferon-alpha genes is lower than anticipated.
  • A significant portion of previously reported gene sequences may represent nonpolymorphic rare mutants, possibly originating from tumor cell lines.
