Summary

A new CD45-SSC gating method improves acute leukemia immunophenotyping by distinguishing leukemic cells from normal cells. This technique enhances accuracy, especially for leukemia with low blast percentages.

Area of Science:

  • Hematology
  • Immunology
  • Cell Biology

Background:

  • Flow cytometry is crucial for immunophenotyping acute leukemia.
  • Current gating relies on Forward Scatter (FSC) and Side Scatter (SSC), which can misidentify normal cells as leukemic.
  • Accurate identification of leukemic cells is vital for diagnosis and treatment.

Purpose of the Study:

  • To develop a more accurate gating method for immunophenotyping acute leukemia.
  • To overcome the limitations of FSC-SSC gating in distinguishing leukemic from normal cells.

Main Methods:

  • Utilized a CD45 monoclonal antibody in conjunction with the SSC parameter for cell gating.
  • Applied the CD45-SSC gating strategy to immunophenotype samples from patients with acute myeloid leukemia (AML) and acute lymphoblastic leukemia (ALL).

Main Results:

  • The CD45-SSC gating method effectively discriminated leukemic cells (CD45dim cells) from contaminating normal cells.
  • This method proved highly beneficial for immunophenotyping AML and ALL, particularly in cases with a low percentage of blasts.

Conclusions:

  • The CD45-SSC gating method offers superior accuracy for immunophenotyping acute leukemia compared to traditional FSC-SSC gating.
  • This novel approach is recommended for routine use in the accurate immunophenotyping of acute leukemia, especially in challenging cases.
