Summary

Cryopreserved normal bone marrow (NBM) stroma can be used in long-term culture (LTC) to support hematopoietic stem cells. This provides a convenient and standardized alternative to fresh NBM stroma for research.

Area of Science:

  • Hematology
  • Stem Cell Biology
  • Biotechnology

Background:

  • Long-term culture (LTC) using normal bone marrow (NBM) stroma is crucial for studying primitive hematopoietic stem cells (LTC-IC).
  • NBM supply can be inconsistent, and stroma requires weeks to mature, limiting experimental availability.
  • Existing efforts to create cell lines have had limited success.

Purpose of the Study:

  • To investigate the utility of cryopreserved NBM and cryopreserved stroma as alternatives to fresh NBM stroma for LTC.
  • To assess the functional capacity of cryopreserved stroma in supporting hematopoiesis.

Main Methods:

  • Comparison of fresh and cryopreserved NBM stroma function in LTC over 12 weeks.
  • Evaluation of cryopreserved stroma's ability to support third-party hematopoiesis in LTC.

Related Experiment Videos

Main Results:

  • Cryopreserved NBM stroma demonstrated similar function to fresh NBM stroma in LTC up to 12 weeks.
  • Stroma from cryopreserved NBM effectively supported third-party hematopoiesis in LTC, comparable to fresh NBM stroma.

Conclusions:

  • Batched cryopreserved stroma offers a convenient, readily available, and standardized source of functional stroma.
  • This advance overcomes logistical challenges and variability associated with fresh NBM stroma.
  • Cryopreserved stroma will facilitate broader application of stroma-based LTC in human hematopoiesis research.
