Background:

  • The role of E-cadherin in renal cell carcinoma (RCC) is unclear, and E-cadherin-negative tumors are more common in RCC than other carcinomas.

Purpose of the Study:

  • To investigate the expression of various cadherins in renal cell carcinoma (RCC) cell lines and normal kidney tissue.
  • To identify cadherin family members beyond E-cadherin that are expressed in RCC.

Main Methods:

  • Immunofluorescence staining for alpha-catenin in 20 RCC cell lines.
  • Immunoprecipitation and Western blotting to detect cadherin-related proteins.
  • RT-PCR with degenerate primers to identify expressed cadherin transcripts.

    • N-cadherin mRNA was ubiquitous, cadherin 6 mRNA was found in 16 lines, cadherin 11 mRNA in five, and E-cadherin protein in six lines.
    • Cadherin 4 transcript was rare, and P-cadherin protein was undetectable. Both normal kidney and RCC tissues expressed multiple cadherins.

    Conclusions:

    • Renal cell carcinoma (RCC) cell lines and tissues express a diverse array of cadherins, including N-cadherin, cadherin 6, and cadherin 11, in addition to E-cadherin.
    • The complex cadherin expression profile in RCC warrants further investigation into their specific roles in tumor behavior and progression.
