[Preparation of apical and basolateral membranes from small intestinal epithelium. Methodologic aspects]
Abstract:
Based on the data from literature and on the authors' results, the methods for isolation of apical and basolateral membranes from small intestinal epithelial cells have been analyzed by the following criteria: medium of isolation, homogenization, scheme of preparation, membranes purity and contamination, orientation of vesicles. It is marked that the process of the tissue homogenization is the important one for all the procedure. Typical localization of membranes in the layers of the gradient of 42.5-48% for apical and of 31-33% for basolateral membranes makes it easy to work out the methods for different subject investigation.
