Summary

Staphylococcal superantigens (SAg) can reduce T cell receptor (TCR) expression by causing internalization of V beta molecules. This SAg-induced TCR modulation may impact infections with Staphylococcus aureus.

Background:

  • Staphylococcal enterotoxins (SAg) are linked to toxic shock syndrome and autoimmune diseases.
  • T cells in these diseases exhibit broad T cell receptor (TCR) V beta region expression.

Purpose of the Study:

  • To investigate the effects of staphylococcal superantigens on TCR V beta cell surface expression.
  • To determine the mechanism of SAg-mediated TCR downregulation.

  • Incubation of peripheral blood mononuclear cells (PBMC) from healthy donors with staphylococcal SAg.
  • Analysis of TCR V beta expression using monoclonal antibodies (mAb).

  • SAg stimulation led to increased intracellular TCR V beta molecules and decreased cell surface expression, indicating TCR internalization.

  • This internalization mechanism explains the observed reduction in cell surface TCR V beta.
