Glomerular hypertrophy, a key factor in FSGS, was observed in the puromycin aminonucleoside (PAN) rat model. Capillary growth initially drives this hypertrophy, followed by changes in diameter and matrix expansion.
Area of Science:
- Nephrology
- Pathology
- Renal Physiology
Background:
- Glomerular hypertrophy is increasingly recognized as a critical factor in focal and segmental glomerulosclerosis (FSGS) development.
- The puromycin aminonucleoside (PAN) rat model has been a standard for FSGS research, though its role in glomerular hypertrophy was previously unclear.
Purpose of the Study:
- To investigate the presence of glomerular hypertrophy in the PAN rat model of FSGS.
- To determine the contribution of glomerular capillary growth to hypertrophy in this model.
Main Methods:
- Female Sprague-Dawley rats were administered PAN or saline subcutaneously over 10 weeks.
- Tissue analysis at 7 and 13 weeks utilized unbiased stereological methods to assess glomerular parameters.
- Statistical analysis included two-way ANOVA to evaluate the effects of PAN and time.
Main Results:
- PAN-treated rats exhibited significant glomerular tuft and renal corpuscle volume increases (48% at 7 weeks, 63% at 13 weeks).
- FSGS was minimal at 13 weeks, indicating hypertrophy precedes significant sclerosis.
- PAN significantly affected capillary length, surface area, and diameter, suggesting capillary remodeling.
Conclusions:
- The PAN model demonstrates a biphasic pattern of glomerular hypertrophy.
- Early hypertrophy (to 7 weeks) is driven by increased capillary length.
- Later enlargement (7-13 weeks) involves increased capillary diameter and/or mesangial matrix expansion.
