Summary

Tryptophan (TRP) depletion in patients with obsessive-compulsive disorder did not worsen symptoms or mood. This study found no significant impact on obsessions, compulsions, or emotional state after TRP reduction.

Background:

  • Obsessive-compulsive disorder (OCD) is a debilitating mental health condition.
  • Tryptophan (TRP) is an essential amino acid precursor to serotonin, a neurotransmitter implicated in mood and behavior.
  • Previous research suggested TRP depletion might exacerbate OCD symptoms.

Purpose of the Study:

  • To investigate the effect of acute tryptophan depletion on obsessive-compulsive symptoms and mood in OCD patients.
  • To examine whether TRP depletion influences symptom severity or emotional state in a controlled clinical setting.

Main Methods:

  • A double-blind, crossover study involving twelve patients diagnosed with OCD.
  • Participants received either a tryptophan-free amino acid mixture or a complete essential amino acid mixture.

  • Plasma TRP levels were monitored, and symptom severity was assessed using Visual Analogue Scales (VAS).
  • Main Results:

    • The tryptophan-free mixture successfully induced significant depletion of plasma TRP.
    • Despite TRP depletion, mean VAS ratings for obsessions and compulsions showed no worsening.
    • Mood assessments also indicated no significant alterations following TRP reduction in this cohort.

    Conclusions:

    • Acute tryptophan depletion does not appear to exacerbate core symptoms of obsessive-compulsive disorder.
    • Contrary to some prior findings, TRP depletion did not negatively impact mood in this patient group.
    • These results suggest that short-term serotonin reduction via TRP depletion may not be a critical factor in OCD symptom expression.
