Summary

This study developed sustained-release microcapsules using cellulose acetate butyrate (CAB) and polystyrene (PS) for ketoprofen delivery. Optimizing the CAB-PS ratio enhanced drug loading and controlled release kinetics, showing potential for improved anti-inflammatory drug formulations.

Area of Science:

  • Materials Science
  • Pharmaceutical Sciences
  • Polymer Chemistry

Background:

  • Development of effective drug delivery systems is crucial for therapeutic efficacy.
  • Microencapsulation offers a method for controlled release of active pharmaceutical ingredients.
  • Ketoprofen, a non-steroidal anti-inflammatory drug, requires formulations for sustained therapeutic action.

Purpose of the Study:

  • To prepare sustained-release microcapsules of ketoprofen using cellulose acetate butyrate (CAB) and polystyrene (PS).
  • To investigate the impact of polystyrene concentration and core/coat ratio on microcapsule properties and drug release.
  • To analyze the drug release kinetics and compare them with release from polymeric films.

Main Methods:

  • Modified W/O/W complex emulsion technique for microencapsulation.

  • Kinetic analysis of ketoprofen release using Higuchi and Baker-Lonsdale models.
  • Preparation and characterization of CAB and CAB-PS free films for permeation studies.
  • Main Results:

    • A CAB:PS ratio of 92.5:7.5 significantly improved microcapsule yield and ketoprofen loading.
    • Drug release from CAB microcapsules followed the Higuchi model, while CAB-PS microcapsules followed the Baker-Lonsdale model.
    • Ketoprofen permeation through CAB-PS matrices was controllable by adjusting the polymer ratio, with sustained release observed from films (58-146 h).

    Conclusions:

    • Polystyrene plays a key role in the manufacturing and release characteristics of CAB microcapsules.
    • The CAB-PS ratio is a critical factor for controlling ketoprofen loading, release rate, and permeation.
    • These microcapsules demonstrate potential for developing sustained-release formulations of ketoprofen.
