Clozapine response and the 5HT2C Cys23Ser polymorphism

Summary

The 5HT2C receptor Ser23 allele does not predict a better response to clozapine treatment. This study found no association between this serotonin receptor variant and clozapine efficacy in patients with psychiatric conditions.

Background:

The 5HT2C receptor Ser23 allele has been suggested to predict a positive response to clozapine, an antipsychotic medication.
This allele, differing from the common Cys23, alters receptor pharmacology, potentially offering insights into antipsychotic drug mechanisms.

Purpose of the Study:

To investigate the association between the 5HT2C receptor Cys23Ser polymorphism and clozapine response in a clinical trial setting.

Main Methods:

Genotyping for the 5HT2C receptor Cys23Ser locus was performed in 66 subjects undergoing double-blind clozapine treatment.
Statistical analyses, including Fisher's exact test and t-tests, were used to assess the relationship between the Ser23 allele and treatment outcomes.

No statistically significant relationship was found between the presence of the Ser23 allele and clozapine response (p = 0.30).
The Ser23 allele did not influence the absolute levels of psychiatric symptoms after 10 weeks of clozapine treatment (p = 0.57).

Conclusions:

The 5HT2C receptor Cys23Ser polymorphism is not associated with clozapine response.
These findings do not support the use of this specific 5HT2C receptor variant as a predictive biomarker for clozapine efficacy.
