Summary

Bipolar disorder patients in manic or hypomanic episodes show hyperpolarized transmembrane potential (TMP) in leukocytes. Euthymic bipolar individuals had normal TMP, suggesting mood states impact cellular membrane physiology.

Area of Science:

  • Neuroscience
  • Cellular Physiology
  • Psychiatry

Background:

  • Ion regulation and distribution abnormalities are frequently observed in bipolar disorder.
  • Altered ion balance may impact cellular physiological functions.

Purpose of the Study:

  • To investigate if ion dysregulation in bipolar disorder affects cellular physiological function.
  • To determine the transmembrane potential (TMP) in leukocytes of bipolar patients during different mood states.

Main Methods:

  • Transmembrane potential (TMP) was measured in mononuclear leukocytes.
  • Flow cytometry utilized the cationic potential-sensitive dye dihexyloxacarbocyanine iodide (DIOC6(3)).
  • Measurements were taken from healthy controls and bipolar patients in normal, manic, and hypomanic phases.

Related Experiment Videos

Main Results:

  • A normal TMP range was established using leukocytes from 5 control individuals.
  • Patients experiencing manic and hypomanic episodes exhibited significantly hyperpolarized TMP (P = 0.0036).
  • Euthymic bipolar individuals displayed TMP levels not significantly different from normal controls.

Conclusions:

  • Pathologic mood states in bipolar illness are associated with altered cellular membrane physiology.
  • TMP changes may serve as a biomarker for mood episodes in bipolar disorder.
