The mu-opioid receptor agonist PL-017 inhibits neuronal activity and alters temperature sensitivity in rat brain slices. This suggests a role for preoptic/anterior hypothalamus neuron temperature sensitivity changes in mu-agonist-induced hyperthermia.

Area of Science:

  • Neuroscience
  • Pharmacology
  • Physiology

Background:

  • The preoptic area/anterior hypothalamus (PO/AH) regulates body temperature.
  • Mu-opioid receptors are implicated in thermoregulation.
  • Selective mu-opioid receptor agonists can induce hyperthermia.

Purpose of the Study:

  • To investigate the effects of the mu-opioid receptor agonist PL-017 on PO/AH neuronal activity and temperature sensitivity.
  • To determine the role of PO/AH neuronal temperature sensitivity in mu-agonist-induced hyperthermia.

Main Methods:

  • Extracellular recordings were performed on rat brain slices.
  • The effects of varying concentrations of PL-017 (a mu-opioid receptor agonist) were examined.
  • The influence of the mu-opioid receptor antagonist CTOP was assessed.

Related Experiment Videos

Main Results:

  • PL-017 inhibited tonic neuronal activity in a dose-dependent manner.
  • Temperature sensitivity was selectively decreased in warm-sensitive neurons at lower concentrations (up to 10 nM).
  • Higher concentrations (100 nM) affected temperature sensitivity in both warm-sensitive and temperature-insensitive neurons.
  • The mu-opioid antagonist CTOP blocked the effects of PL-017.

Conclusions:

  • Mu-opioid receptor activation in the PO/AH alters neuronal tonic activity and temperature sensitivity.
  • Selective changes in PO/AH neuron temperature sensitivity are implicated in mu-agonist-induced hyperthermia.
