Purpose of the Study:

  • To investigate the impact of aniso-osmolarity on Erk-1 and Erk-2 activity in C6 glioma cells.

  • C6 glioma cells were subjected to hypo-osmotic and hyper-osmotic conditions.
  • Erk-1 and Erk-2 activity was measured using mobility shifts and activity assays.
  • Involvement of calcium, phospholipase C, Ras/Raf pathway, protein kinase C, G-proteins, tyrosine kinases, and PI3 kinase was assessed using specific inhibitors.

  • The osmosignaling pathway upstream of MAP kinases in C6 glioma cells is distinct from that in primary rat astrocytes, H4IIE rat hepatoma cells, and isolated rat hepatocytes.
  • These findings highlight cell-type-specific differences in cellular responses to osmotic challenges.
