    New rat testicular perfusion methods allow direct testosterone measurement. The isolated testis method is more sensitive but cannot maintain energy levels for high-energy phosphate metabolism studies.

    Background:

    • Organ perfusion techniques are valuable for biological studies but need thorough validation.
    • Rat testicular perfusion is crucial for understanding reproductive functions and hormone secretion.

    Purpose of the Study:

    • To characterize and compare two novel nonrecirculating rat testicular perfusion methods: selective and isolated testicular perfusion.
    • To assess the suitability of these methods for studying testosterone secretion and related biological processes.

    Main Methods:

    • Characterization of selective and isolated testicular perfusion systems in rats.
    • Comparison with a recirculating hemicorpus system.
    • Measurement of testosterone production in response to varying doses of human chorionic gonadotropin (hCG).

    Main Results:

    • Both methods allowed direct measurement of testosterone secretion, which was dose-dependent on hCG up to 1000 mIU/mL.
    • The isolated testis method demonstrated higher sensitivity to hCG, required less medium, and maintained better physiological parameters (flow rate, water content, no initial ischemia) compared to the selective method.
    • Neither method sustained normal ATP and GTP levels after 3 hours of perfusion.

    Conclusions:

    • Both selective and isolated testicular perfusion methods are suitable for studying testosterone secretion and androgen receptors.
    • The inability of current methods to maintain testicular ATP and GTP levels limits their application in studies of high-energy phosphate metabolism.
