The indirect rat mast cell degranulation test reconsidered

P G Krüger, J Apold, S Elsayed

    Summary

The indirect rat mast cell degranulation test is unreliable for detecting human reaginic antibodies. High serum concentrations cause non-specific histamine release, invalidating allergen-specific results in immediate hypersensitivity studies.

    Area of Science:

    • Immunology
    • Allergy Research

    Background:

    • The indirect rat mast cell degranulation test has been explored for detecting human reaginic antibodies.
    • Immediate hypersensitivity reactions involve mast cell degranulation and histamine release.

    Purpose of the Study:

    • To re-evaluate the validity of the indirect rat mast cell degranulation test for detecting human reaginic antibodies.
    • To determine the influence of serum concentration and allergen presence on histamine release in this test system.

    Main Methods:

    • Incubation of rat mast cells with varying concentrations of human reaginic serum.
    • Assessment of histamine release in response to different serum concentrations and allergen exposure.

    Main Results:

    • A massive, non-specific histamine release was observed when serum concentrations exceeded approximately 3%.
    • At tolerated serum concentrations (below 3%), allergen concentration did not significantly affect histamine release.

    Conclusions:

    • The indirect rat mast cell degranulation test is not a suitable alternative for studying immediate hypersensitivity in humans.
    • The test's sensitivity to non-specific histamine release at higher serum concentrations compromises its diagnostic utility.
