Area of Science:

  • Thoracic Surgery
  • Pulmonary Medicine
  • Surgical Models

Background:

  • Established a rat model for isolated single-lung perfusion to study chemotherapy for pulmonary metastases.
  • Designed a sequential bilateral isolated lung perfusion model to assess staged perfusions in rats.

Purpose of the Study:

  • To investigate the feasibility and safety of sequential bilateral isolated lung perfusion in a rat model.
  • To establish a reliable model for future studies on chemotherapeutic agents for bilateral pulmonary metastases.

Main Methods:

  • Twenty-four Fischer rats were divided into three experimental groups (n=8) undergoing left isolated lung perfusion, followed by contralateral perfusion at 1, 2, or 3 weeks.
  • A control group (n=5) underwent sequential bilateral sham thoracotomies.

Related Experiment Videos

  • Arterial blood gas analysis was performed post-second operation in one experimental group and the control group.
  • Main Results:

    • Survival rates for the second operation were 100% (Group I), 75% (Group II), and 100% (Group III). All control animals survived.
    • Arterial blood gas analysis showed no significant differences in oxygen, carbon dioxide, or pH between the experimental and control groups.

    Conclusions:

    • Sequential bilateral isolated lung perfusion is safe and well-tolerated in rats.
    • This model is suitable for evaluating the safety and efficacy of staged perfusions for treating bilateral pulmonary metastases.
