Summary

NMDA receptor antagonists altered dopamine neuron firing patterns in rats. While these drugs reduced bursting activity, they did not eliminate it, suggesting NMDA receptors are not solely responsible for dopamine neuron bursting.

Background:

  • Mesencephalic dopamine (DA) neurons modulate neural circuits through their firing patterns.
  • Activity patterns of DA neurons are thought to depend on N-methyl-D-aspartate (NMDA) receptor activation.

Purpose of the Study:

  • To investigate the effects of NMDA receptor antagonists on the firing properties of nigral DA neurons in rats.
  • To determine the role of tonic NMDA receptor activation in DA neuronal bursting activity.

Main Methods:

  • Extracellular single-unit recordings were performed in anesthetized rats.
  • The effects of four NMDA receptor antagonists (CGS-19755, (+/-)-CPP, NPC-12626, NPC-17742) on DA neuron firing were assessed.

Main Results:

  • All tested NMDA receptor antagonists increased DA neuron firing rate and regularized firing patterns.
