Science (New York, N.Y.)·2024

Area of Science:

  • Pharmaceutical technology
  • Respiratory medicine
  • Medical device engineering

Background:

  • Efficient aerosol drug delivery to the lungs is crucial for effective pulmonary and systemic therapies.
  • Existing inhalation devices face challenges in controlling aerosol properties for consistent dosing.

Purpose of the Study:

  • To characterize the in vitro performance of the AERX aerosol delivery system.
  • To evaluate its suitability for pharmacokinetic and scintigraphic studies.

Main Methods:

  • The AERX system, a bolus inhaler, was tested using liquid formulations in collapsible containers.
  • Aerosol generation involved forcing formulation through 2.5-micron holes at 70 LPM.
  • Particle size distribution and emitted dose were quantified using filters and an Andersen cascade impactor.

Main Results:

  • The system emitted 57.0% +/- 5.9% of the loaded drug dose as aerosol.
  • The aerosol particle size distribution showed a Mass Median Aerodynamic Diameter (MMAD) of 2.95 +/- 0.06 microns.
  • A narrow particle size distribution was observed with a geometric standard deviation (sigma g) of 1.24 +/- 0.01.

Conclusions:

  • The AERX system reliably produces a nearly monodisperse aerosol.
  • The generated aerosol possesses characteristics suitable for efficient and reproducible lung drug delivery.
