Summary

Staphylococcal enterotoxin B (SEB) depletes V beta 8+ T-cells in mice. A novel V beta 8+CD4-CD8- T-cell subpopulation emerges, potentially autoreactive.

Area of Science:

  • Immunology
  • T-cell biology

Background:

  • Staphylococcal enterotoxin B (SEB) is a superantigen known to activate T-cells.
  • SEB triggers massive T-cell activation and subsequent deletion of specific T-cell receptor (TCR) V beta elements.

Purpose of the Study:

  • To investigate the impact of SEB injection on T-cell populations in BALB/c mice.
  • To characterize the changes in V beta 8+ T-cells, including CD4 and CD8 expression.
  • To identify and analyze any novel T-cell subpopulations induced by SEB.

Main Methods:

  • Injection of SEB into BALB/c mice.
  • Flow cytometry analysis to assess T-cell populations (V beta 8+, CD4+, CD8+).
  • Assessment of cell surface molecule expression at 72 hours post-injection.
  • In vitro proliferation assays using syngeneic splenocytes.

Related Experiment Videos

Main Results:

  • SEB induced depletion of V beta 8+ T-cells, affecting both V beta 8+CD4+ and V beta 8+CD8+ subsets.
  • CD8 molecule expression decreased on V beta 8+CD8+ cells at 72 hours post-SEB.
  • A distinct V beta 8+CD4-CD8- T-cell subpopulation was induced in spleen and lymph nodes.
  • This V beta 8+CD4-CD8- subpopulation constituted a major portion of V beta 8+ cells in lymph nodes at 72 hours.
  • Some V beta 8+CD4-CD8- T-cells exhibited proliferative responses to syngeneic splenocytes, suggesting potential autoreactivity.

Conclusions:

  • SEB triggers selective depletion of specific T-cell subsets.
  • SEB induces a novel V beta 8+CD4-CD8- T-cell population.
  • This newly identified T-cell subpopulation may possess autoreactive properties.
