Purpose of the Study:

  • To investigate the relationship between P1/P2 ratios and standard semen parameters.

Main Methods:

  • Sperm samples from male-factor infertility patients undergoing IVF were analyzed.
  • Protamines were extracted and separated using acid urea polyacrylamide gel electrophoresis.
  • Nonparametric analysis was used to correlate P1/P2 ratios with IVF outcomes and semen parameters.

Main Results:

  • The P1/P2 ratio in a control group with successful IVF (>50% fertilization) ranged from 0.55 to 1.29.
  • Patients with lower IVF rates (<50%) showed P1/P2 ratios outside the control range, with some exhibiting abnormally large sperm heads.
  • A negative correlation was observed between P1/P2 ratios and IVF rates in the control group.

Conclusions:

  • P1/P2 ratios showed a negative correlation with IVF rates and progressive motile sperm concentration.
  • While altered P1/P2 ratios were noted in some infertile men, they are unlikely to be the primary cause of reduced fertilization.
  • Further research is needed to fully elucidate the role of protamine ratios in male fertility.
