Purpose of the Study:

  • To investigate the impact of chronic morphine on hypothalamic oxytocin levels.
  • To explore oxytocin neuron function in opioid tolerance.

Main Methods:

  • Male rats received chronic morphine or placebo pellets for 7 days.
  • Acute saline or morphine injections were administered.
  • Oxytocin content in hypothalamic nuclei was measured.

Main Results:

  • Morphine tolerance decreased oxytocin in the median eminence and supraoptic nucleus.
  • No changes were observed in the paraventricular nucleus.
  • These alterations occurred after both saline and morphine challenges.

Conclusions:

  • Chronic morphine exposure modifies the brain oxytocin system.
  • Oxytocin may play a role in opioid-induced behavioral and neuroendocrine effects.
