Background:

  • Transient occlusion of carotid arteries causes retinal ischemia, reducing the electroretinogram (ERG) b-wave amplitude.
  • Oxidative stress is implicated in ischemic retinal dysfunction.

Purpose of the Study:

  • To investigate the protective effects of vitamin E and lipoate against retinal ischemic dysfunction.
  • To determine if these antioxidants attenuate b-wave depression and enhance recovery during retinal ischemia.

Main Methods:

  • Rats underwent transient (24 min) bilateral common carotid artery occlusion under halothane anesthesia.
  • Electroretinograms (ERGs) were recorded before, during, and after ischemia.
  • Vitamin E and lipoate were administered 20 minutes before or at the onset of ischemia.

Main Results:

  • Both vitamin E and lipoate significantly reduced the depression of the ERG b-wave during occlusion.
  • These antioxidants accelerated the recovery of the b-wave during reperfusion.
  • Protective effects were observed regardless of administration timing (pre-ischemia or onset).

Conclusions:

  • Antioxidants, specifically vitamin E and lipoate, offer protection against functional impairment of the retina during ischemia.
  • These findings suggest a therapeutic potential for antioxidants in managing ischemic retinal conditions.
