Summary

Cyclin D1, a proto-oncogene, is frequently overexpressed in mantle cell lymphoma due to chromosomal translocations. This overexpression serves as a sensitive and specific diagnostic marker for MCL.

Area of Science:

  • Oncology
  • Molecular Biology
  • Hematology

Background:

  • Cyclin D1 is a proto-oncogene regulating cell cycle progression.
  • Its deregulation is implicated in tumor development and uncontrolled cell growth.
  • The chromosomal translocation t(11;14)(q13:q32) is linked to lymphoid neoplasia, specifically mantle cell lymphoma (MCL).

Purpose of the Study:

  • To review current knowledge on cyclin D1 overexpression in malignant lymphomas.
  • To emphasize the clinicopathologic significance of cyclin D1 in these cancers.
  • To highlight cyclin D1 as a potential diagnostic marker for MCL.

Main Methods:

  • Review of recent studies on cyclin D1 in lymphomas.
  • Analysis of immunohistochemical detection methods for cyclin D1.
  • Correlation of cyclin D1 expression with clinicopathologic features.

Main Results:

  • The BCL-1 locus rearrangement, involving the cyclin D1 gene, occurs in 40-70% of MCL cases.
  • Break points in BCL-1 maintain the cyclin D1 coding region, leading to increased protein expression.
  • Immunohistochemistry using monoclonal antibodies on paraffin-embedded tissue is effective for routine diagnosis.

Conclusions:

  • Cyclin D1 overexpression is a hallmark of MCL, distinct from other B cell lymphomas.
  • Increased cyclin D1 protein expression is a sensitive and specific marker for MCL diagnosis.
  • Immunohistochemical detection of cyclin D1 is a valuable tool in routine diagnostic settings.
