Area of Science:

  • Obstetrics and Gynecology
  • Pharmacology
  • Teratology

Background:

  • Metronidazole is a commonly prescribed antibiotic during pregnancy.
  • Concerns exist regarding potential teratogenic effects of metronidazole.
  • Evidence on the association between metronidazole and congenital malformations is inconclusive.

Purpose of the Study:

  • To evaluate the risk of congenital malformations associated with metronidazole use during pregnancy.
  • To synthesize existing epidemiological data through a meta-analysis.

Main Methods:

  • Systematic review and meta-analysis of epidemiological studies (cohort and case-control).
  • Inclusion of studies assessing congenital malformation risk after first-trimester metronidazole exposure.
  • Application of the Mantel-Haenszel method to calculate summary odds ratio.

Related Experiment Videos

  • Heterogeneity testing to assess study consistency.
  • Main Results:

    • Five studies (1 unpublished case-control, 4 published cohort) met inclusion criteria.
    • Studies were not statistically heterogeneous.
    • Summary odds ratio for metronidazole exposure in the first trimester was 1.08 (95% CI: 0.90-1.29).
    • Heterogeneity test: chi2 = 4.72, P = 0.32.

    Conclusions:

    • This meta-analysis did not identify a relationship between first-trimester metronidazole exposure and birth defects.
    • The findings suggest that metronidazole use during early pregnancy is not associated with an increased risk of congenital malformations.
