Summary

Subcutaneous cisplatin in dogs showed toxicity and slow systemic absorption. A drug delivery vehicle may be needed for sustained release and reduced adverse effects in cancer treatment.

Area of Science:

  • Veterinary Medicine
  • Pharmacology
  • Oncology

Background:

  • Previous polymer release studies informed this investigation.
  • Evaluating novel drug delivery for cancer therapy is crucial.

Purpose of the Study:

  • To assess the toxicity, efficacy, and systemic absorption of subcutaneous cisplatin in dogs.
  • To determine the feasibility of using subcutaneous cisplatin as an adjunct cancer treatment.

Main Methods:

  • Six dogs with sarcoma received 50-70 mg/m2 cisplatin suspended in saline via subcutaneous injection.
  • Toxicity, serum platinum levels, and clinical outcomes were monitored post-administration.

Main Results:

  • Significant gastrointestinal, bone marrow, renal, and local tissue toxicity were observed.

Related Experiment Videos

  • Measurable serum platinum levels persisted up to 14 days post-injection.
  • Treatments were discontinued due to toxicity, with dogs completing adjuvant therapy using intravenous carboplatin.
  • Conclusions:

    • Subcutaneous cisplatin administration in dogs resulted in considerable toxicity.
    • Slow systemic absorption of platinum occurred, but was associated with adverse effects.
    • A specialized drug delivery vehicle may be required for safe and effective sustained-release subcutaneous cisplatin therapy.
