Summary

Basic fibroblast growth factor (FGF2) can acutely trigger seizures and alter hippocampal electrical activity in rats. This finding reveals a novel, rapid effect of FGF2 beyond its known neurotrophic roles.

Background:

  • Basic fibroblast growth factor (FGF2) is known for its neurotrophic effects on hippocampal neurons.
  • The acute impact of FGF2 on hippocampal electrical activity in vivo remains uninvestigated.

Purpose of the Study:

  • To investigate the acute effects of FGF2 on hippocampal electroencephalographic (EEG) activity and behavior in freely moving animals.

Main Methods:

  • Two doses of FGF2 (50 ng and 25 ng) were injected into the ventral hippocampus of rats.
  • EEG recordings and behavioral observations were used to monitor seizure activity.

Main Results:

  • A 50 ng dose of FGF2 induced immediate EEG ictal discharges and behavioral seizures lasting approximately 2 hours in 7/9 rats.
  • A 25 ng dose of FGF2 induced seizures of about 60 minutes in 3/7 rats.

  • Heat-inactivated FGF2 did not produce any significant EEG or behavioral changes.
