Background:

  • Major depressive disorder (MDD) is often associated with altered serotonergic (5-HT) neurotransmission.
  • Serotonergic neuroendocrine challenges are used to assess 5-HT system function in MDD.

Purpose of the Study:

  • To investigate the effects of antidepressant treatment on serotonergic function in patients with unipolar major depression.
  • To assess neuroendocrine responses to d-fenfluramine before and after treatment with a noradrenergic reuptake inhibitor, tricyclic antidepressant, or placebo.

Main Methods:

  • A serotonergic neuroendocrine challenge using d-fenfluramine (30 mg) was administered to patients with MDD at baseline.
  • Patients received 6 weeks of treatment with a noradrenergic reuptake inhibitor, tricyclic antidepressant, or placebo.
